Twenty-two people. That is the entire human evidence base for the most-cited “brain strain” on the internet.

I went looking for this because the strain codes impressed me. B. longum 1714. TWK10. BC30. Those numbers signal precision, and precision usually means somebody ran a proper trial. Somebody did. Then I opened the papers and found sample sizes you could fit in a classroom.

The strains are real. The trials are real. The gap between what they measured and what gets promised is where this gets interesting.

This one has the best story attached to it, so it deserves the closest look.

Researchers at University College Cork gave healthy volunteers the 1714 strain, then stressed them on purpose with a cold pressor test while people watched. Cortisol output rose less than it did on placebo. Self-reported daily stress dropped. Visuospatial memory improved slightly, and resting brain activity shifted (B. longum 1714 as a translational psychobiotic — Translational Psychiatry).

That is a genuine result, and I want to be fair to it. The design was careful. The measurements were objective, not just questionnaires.

Twenty-two people took part. The researchers describe the benefits in their own paper as subtle and modest. One of the co-authors was affiliated with the company that makes the strain.

A 2024 randomised trial later looked at sleep quality and wellbeing with the same strain (B. longum 1714 and sleep quality — Scientific Reports). The field is building. It has not arrived.

“Heals your brain” is not what twenty-two people and a cold water bath established.

The thread claims most probiotics die in digestion. That is roughly right for many products, and BC30 is the reason the claim needs an asterisk.

B. coagulans BC30 forms spores. Spores are dormant, armoured, and built to survive acid. In a simulated gut model, survival came in around 70 percent, after which it germinates in the small intestine (BC30 protein absorption review).

The human trial gave thirty people milk protein with or without BC30. Blood amino acid peaks came in higher and faster with the strain added (BC30 and amino acid absorption — Nutrition & Metabolism).

So BC30 has a defensible mechanism and a measurable effect. The effect is on protein absorption. The thread files it under metabolism and muscle, which quietly upgrades “your amino acids showed up in the blood sooner” into something about body composition. Nobody measured that.

Worth knowing: BC30 is a patented commercial strain. The research is largely funded by companies that sell it.

TWK10 has more human trials than most strains in the thread. The first had sixteen people, split eight and eight (TWK10 endurance trial). A later one on the heat-killed form had thirty (heat-killed TWK10 — Physiological Reports).
