When most people hear the word melatonin, they think of one thing:
Sleep.
A supplement taken at night.
A signal that tells the body it is time to rest.
But this is only one small piece of the story.
Melatonin is one of the most fascinating molecules in human biology.
It is involved in:
• sleep and circadian rhythm
• mitochondrial function
• antioxidant protection
• immune regulation
• aging biology
• cellular repair
And perhaps most importantly:
Melatonin is part of the conversation between light and darkness.
Because the story of melatonin doesn’t begin with a pill.
It begins with sunlight.
The Sunlight-Darkness Connection
At first, it seems strange that sunlight would be connected to a hormone produced in darkness.
But this is exactly how biology works.
The body does not experience light and darkness as separate events.
They are part of one continuous rhythm.
Morning sunlight tells your brain:
“The day has begun.”
This initiates a cascade of signals that eventually allows your body to create:
“Night is coming. Prepare for repair.”
Without strong daytime light signals, the body often struggles to create a strong nighttime melatonin response.
You make melatonin in UVA morning sunlight as tryptophan get converted into serotonin, which later becomes melatonin in the absence of light at night.
Light builds the rhythm.
Darkness activates the hormone.
Melatonin Is Not Just a Sleep Hormone
Melatonin is produced primarily by the pineal gland at night.
As light decreases, the brain receives the signal that it is time to transition into darkness. It takes 4 hours of darkness for melatonin to be secreted. Let that sink in … It could be peaking around midnight, but what if you're scrolling on your phone or watching TV late into the evening…. that melatonin does not peak…
The pineal gland begins producing melatonin, which helps coordinate:
• sleep timing
• body temperature changes
• metabolic processes
• tissue repair
But melatonin is not simply a “sleep chemical.”
It is a powerful signaling molecule with effects throughout the entire body.
The Mitochondrial Melatonin Connection
One of the most exciting areas of melatonin research involves the mitochondria.
Most people think of mitochondria only as energy producers.
But mitochondria are also one of the primary places where the body experiences oxidative stress.
Every time mitochondria create energy, they naturally produce reactive oxygen species (ROS).
This is normal.
The problem occurs when oxidative stress exceeds the body’s ability to regulate and repair.
This is where melatonin becomes fascinating.
Research suggests that melatonin is produced inside mitochondria themselves, where it may act as a local antioxidant and help protect these energy-producing structures.
This mitochondria melatonin is 95% of our melatonin production and is made in response to near infrared light, which is available from the sun all day long even in shade.
Mitochondrial melatonin helps:
• support cellular resilience
• regulate oxidative stress
• protect mitochondrial DNA
• support efficient energy production
In other words:
Melatonin isn’t just helping you sleep.
It helps your cells maintain themselves.
Why Darkness Matters
Modern life has changed our relationship with darkness.
For most of human history, night meant:
• darkness
• lower temperatures
• rest
• restoration
Today, many people experience:
• bright LED lighting at night
• televisions in bedrooms
• phones beside the bed
• late-night computer work
• constant artificial stimulation
The body receives a confusing message:
The clock says nighttime.
The environment says daytime.
This can interfere with normal melatonin production.
Blue Light Is Only Part of the Story
Most conversations focus on blue light.
And blue light does matter.
The specialized light receptors in the eye are especially sensitive to blue wavelengths, which strongly influence circadian timing.
But the bigger issue is not one color of light.
It is the mismatch between our modern environment and our biology.
Our ancestors experienced:
☀️ bright full-spectrum light during the day
🌅 changing colors at sunset
🌙 true darkness at night
Our modern environment often provides:
💡 dim artificial light all day
📱 bright screens at night
This creates a completely different biological signal.

Melatonin and Immune Function
Melatonin also plays an important role in immune regulation.
The immune system is deeply connected to circadian rhythms.
Our bodies have different immune priorities during the day versus at night.
Melatonin helps coordinate these rhythms.
Research has explored melatonin’s role in:
• regulating inflammatory pathways
• supporting antioxidant defenses
• influencing immune cell activity
This is another reminder that sleep is not passive.
Nighttime is an active biological process.
Melatonin and Cancer Research
One of the most fascinating areas of melatonin research is its relationship with cancer biology.
Researchers have investigated melatonin’s potential roles in:
• reducing oxidative stress
• supporting healthy cellular signaling
• influencing hormone pathways
• regulating immune responses
This is especially interesting in hormone-sensitive cancers because melatonin interacts with endocrine signaling.
For example, melatonin has been studied in relation to estrogen pathways and breast cancer biology.
Maintaining healthy circadian rhythms, adequate nighttime darkness, and strong natural melatonin production is an important piece of supporting overall health.
The Melatonin-Vitamin D Relationship
The sunlight story is incomplete if we only talk about vitamin D.
During the day:
Sunlight signals wakefulness.

You cannot optimize nighttime biology without respecting daytime biology.
Why Many People Are Melatonin Deficient
The problem isn’t necessarily that the body cannot make melatonin.
Often, the problem is that we are not giving the body the correct signals.
Common disruptions include:
• not getting morning sunlight
• spending all day indoors
• excessive artificial light at night
• irregular sleep schedules
• chronic stress
• eating late into the night
The solution is not always taking a melatonin supplement.
Sometimes the solution is rebuilding the environment that tells your body when to make it.

The most powerful strategies are often the simplest:

Get outside early in the day. Sunrise and UVA without sunglasses
This strengthens your circadian rhythm and creates a stronger contrast between daytime and nighttime.
Bright Days, Dim Nights
Your body needs a difference between light and darkness.
Bright days.
Dark nights.
This contrast is one of the strongest biological signals we have.
